Treatment protocol |
Exp1: cells were treated with doxycycline at 100 ng/mL for 72 hours to induce GRP128. Exp2A: cells were treated with doxycycline at 500 ng/mL for 24 hours. Exp2B: cells were treated with doxycycline at 500 ng/mL for 96 hours. Cells transfected with only lenti-vector were used as a negative control.

TM-1 cells were seeded at 10'5/mL in the wells of 6-well plates. Cells were maintained in reduced serum (2.5%)

Data processing |
Image data were processed with Agilent Feature Extraction 11.0.1.1, and processed signals were quantile normalized with Agilent GeneSpring 12.0. The normalized expression intensities for all probes were subjected to a two-way analysis of variance analysis (ANOVA). Significantly differentially expressed genes were selected using criteria p-value (or FDR using the Benjimini-Hochberg method) < 0.05 and fold-change > 1.2.
