Sample GSM7516825 Query DataSets for GSM7516825
Status Public on Jun 30, 2024
Title EXP1-rep2-GRP158-72 H
Sample type RNA

Source name human trabecular meshwork (HTM) cells.
Organism Homo sapiens
Characteristics cell line: HTM cells transfected with TIGR/MYOC cDNA (TM-1).
cell type: human trabecular meshwork (HTM) cells
treatment: GRP158-72 H
Treatment protocol Exp1: cells were treated with doxycycline at 100 ng/mL for 72 hours to induce GRP128. Exp2A: cells were treated with doxycycline at 500 ng/mL for 24 hours. Exp2B: cells were treated with doxycycline at 500 ng/mL for 96 hours.
Cells transfected with only lenti-vector were used as a negative control.
Growth protocol TM-1 cells were seeded at 10'5/mL in the wells of 6-well plates. Cells were maintained in reduced serum (2.5%)
Extracted molecule total RNA
Extraction protocol Total RNA was extracted from each culture plate well using Aurum (Bio-Rad, Hercules, CA)
Label Cy3
Label protocol cRNA was labelled with Cyanine-3

Hybridization protocol Cyanine-3-labelled cRNA was hybridized on Agilent SurePrint G3 Human GE 8×60K Microarray v2.
Scan protocol Arrays were scanned with an Agilent DNA Microarray Scanner at a 3-um scan resolution
Description replicate 2
Data processing Image data were processed with Agilent Feature Extraction 11.0.1.1, and processed signals were quantile normalized with Agilent GeneSpring 12.0.
The normalized expression intensities for all probes were subjected to a two-way analysis of variance analysis (ANOVA).  Significantly differentially expressed genes were selected using criteria p-value (or FDR using the Benjimini-Hochberg method) < 0.05 and fold-change > 1.2.
