Sample GSM749431 Query DataSets for GSM749431
Status Public on Dec 21, 2012
Title FABP4-cre-HighFatDiet-rep2
Sample type RNA

Source name White adipose tissue dissected from mice that with adipocyte-specific deletion of Sirt1 on a high fat diet for 8 weeks
Organism Mus musculus
Characteristics background strain: C57BL6/J
cre transgene: Fatty acid binding protein 4 promoter-Cre (FABP4-Cre)
diet: High Fat
Treatment protocol White adipose tissue dissected from mice andimmediately processed immediately to isolate RNA
Growth protocol 12 week old mice with indicated genotypes fed high or low fat diets for 8 weeks-feeding started at 4 weeks of age
Extracted molecule total RNA
Extraction protocol Trizol extraction of total RNA was performed according to the manufacturer's instructions. RNA was DNAseI treated and further purified using the QIAgen Rneasy mini columns
Label Biotin
Label protocol Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 5 ug total RNA according to the Affymetrix Expression Analysis Technical Manual.

Hybridization protocol Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Drosophila Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.
Scan protocol GeneChips were scanned using the GeneChip Scanner 3000.
Description Gene expression data from mouse white adipose tissue without Sirt1 fed a high fat diet
Data processing R version 2.11.1 (x86_64-pc-linux-gnu), affyPLM_1.24.1, preprocessCore_1.10.0, gcrma_2.20.0, affy_1.26.1, Biobase_2.8.0
