The isolation and serum-free culture conditions for the HRTPT cells has been previously described.10, 11 Confluent cultures of HRTPT cells were exposed to 4.5 µM iAS for 24 hrs and then subcultured at a 1:3 ratio in the continued presence of iAS until confluent. Following confluence, the cells were serially sub cultured again in the present of iAS until confluent. This was repeated for 10 serial passages. Additional cultures of iAS-exposed cells at passage 8 were subcultured into iAS-free growth media and continued in iAS-free media for 11 additional passages.

Quality of supplied RNA was checked using the nanodrop (Table 2) and determined to be of enough quality and quantity to proceed with processing onto the Clariom™ D microarray (Table 2). Samples were hybridized on to the Clariom™ D array using the GeneChip™ WT PLUS reagent kit as per the manufacturers protocol. Arrays were scanned using a GeneChip™ GCS 3000 scanner with autoloader. Data analysis was completed using Affymetrix Transcript Analysis console (TAC) and custom bioinformatic pipelines developed at Genome Explorations. All 18 Clariom™ D microarrays passed the internal QC for running the arrays (Table 2). All samples were run together as a single batch to avoid potential batch effects.
