Extraction protocol |
Total RNA extraction using Qiagen's RNeasy Plus Mini Kit was performed according to the manufacturer's instructions. RNA was quantified using a NanoDrop spectrophotometer and quality was monitored with the Agilent 2100 Bioanalyzer.

The sense cDNA was then fragmented and biotin-labeled with TdT (terminal deoxynucleotidyl transferase) using the GeneChip WT Terminal labeling kit.

Hybridization protocol |
Following fragmentation, 5.5 ug of cDNA were hybridized for 16 hr at 45C on GeneChip Drosophila Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.

Scan protocol |
Hybridized arrays were washed and stained on the Agilent Technologies GeneChip Fluidics Station 450 and scanned on the Agilent Technologies Scanner GCS3000.
