Chondrocytes are cultured in 1% FBS, glutamine, ascorbic acid and proline with or without CL and BS for 6h or 24h

Primary OA chondrocytes were isolated from cartilage after enzymatic digestion and growth in alginate beads in high glucose DMEM containing 1% FBS, 2mM glutamine, 50 µg/ml ascorbic acid and 20 µg/ml proline during 3 days

18 beads were pooled, degelified in 0,1M cold citrate and cells were homogenized in 600 µl RTL lysis buffer (Qiagen) then extract using Rneasy mini kit from Qiagen according to the manufacturer's instructions. Dnase step were performed during extraction. Samples were eluted in 30 µl RNase free H2O 0,1 µg of RNA from each culture condition was used for this analysis. Libraries were prepared with the Illumina Truseq stranded mRNA sample prep kit according to the manufacturer's instructions

Data processing |
Raw data were demultiplexed into individual libraries. After filtering out reads mapping to rRNA, tRNA, mitochondrial RNA, and other contaminants (e.g., adapters, etc.) using bowtie2 Assembly: GRCh38 Supplementary files format and content: reads were aligned onto the human reference genome (GRCh38 and Ensembl 105 annotation) and quantified with Star to give the Counts file.
