Growth protocol mouse E12.5 fetal liver cells were expanded in vitro for 3 days in the presence of growth factors then ter119 purified. Ter119- populations were processed for ChIP experiments.

Extraction protocol Protein-DNA cross-linking was performed on 1 x 10^7 Ter119– cells with 1% formaldehyde for 10 min at room temperature. Glycine (0.125 M) was added to quench the reaction. Cells were washed twice in phosphate-buffered saline containing protease inhibitors (Complete; Roche). Nuclei were lysed in lysis buffer (ChIP assay kit; Upstate Biotechnology) and sheared by sonication to reduce the chromatin fragments to 200 to 500 base pairs. Immunoprecipitation of cross-linked chromatin was performed using anti-SCL and ready-made buffers (ChIP assay kit; Upstate Biotechnology). Control samples were treated exactly as the anti-SCL ChIP smaples except no antibody was added to the crosslinked and sonicated chromatin.
