Growth protocol Pre-cultures of SG200-deltahxt1 were grown overnight in liquid array medium (6.25 % (w/v) Holiday salt solution, 30 mM L-glutamine, pH 7.0, filter sterilized) containing 1% glucose (w/v). SG200-deltahxt1 cells were shifted to fresh liquid array medium containing 1% glucose and incubated for 6 hours to an OD600 of 1.0. RNA samples were frozen in liquid nitrogen for two independently conducted replicates.
Extracted molecule total RNA
Extraction protocol Total RNA was extracted using the TRIZOL-reagent (Invitrogen) according to the manufacturer`s instructions. RNA samples were further column purified (Qiagen RNeasy) and the quality checked using a Bioanalyzer with a RNA 6000 Nano LabChip kit (Agilent).
Label biotin/phycoerythrin
Label protocol Target preparation was performed according to the standard Affymetrix protocol using the Enzo BioArray RNA transcript labeling kit, exept for a reaction temperature of 50°C for first-strand cDNA synthesis.
