Growth protocol |
Pre-cultures of SG200-deltahxt1 were grown overnight in liquid array medium (6.25 % (w/v) Holiday salt solution, 30 mM L-glutamine, pH 7.0, filter sterilized) containing 1% glucose (w/v). SG200-deltahxt1 cells were shifted to fresh liquid array medium containing 1% glucose and incubated for 6 hours to an OD600 of 1.0. RNA samples were frozen in liquid nitrogen for two independently conducted replicates.

Extraction protocol |
Total RNA was extracted using the TRIZOL-reagent (Invitrogen) according to the manufacturer`s instructions. RNA samples were further column purified (Qiagen RNeasy) and the quality checked using a Bioanalyzer with a RNA 6000 Nano LabChip kit (Agilent).
