Around 50 embryos collected within 3 hours were put in an individual vial of fly food to avoid crowding and the larvae were raised at 25-degree incubator for appropriate lengths of time before dissection.

The wing imaginal discs were dissociated in 0.25% Trypsin-EDTA solution at 37 ℃ for 10 min. Cells were then washed in DPBS and passed through 35μm filter before library preparation. Construction of 10x single cell libraries and sequencing on Illumina Hiseq platform were performed by Novogene.

Data processing |
Raw data mapping and primary analysis was performed in the Cell Ranger pipeline. The first 26 bp of R1 contains the barcode and UMI. Secondary analysis for marker gene expression pattern was performed with Seurat (Butler et al., 2018). Reads were normalized with the log normalization function in Seurat. Briefly: for each cell, sum all UMI number across all genes, this number is S; for each gene, the normalisation result = log1p(UMI number (of this gene)/S).
