Resting biopsies were taken from the vastus lateralis muscle using the Bergstroem technique.
Tissue was snap frozen in liquid nitrogen-cooled isopentane and kept frozen at -70 deg C.

total RNA was isolated from 25 micrometer cryosections of the muscle biopsies using RNeasy kit (Qiagen).
2.5 micogram total RNA was reverse-transcribed using Superscript II (Invitrogen, Basel, Switzerland).

Standardized amounts of [alpha-32P-dCTP] were used per reaction. Radiolabelled cDNA target was purified from the reaction using the supplied cartridge (BD Biosciences).

Equal amounts (1-3 Mio cpm) of cDNA target were hybridized to individual cDNA array filters in Expresshyp (BD Biosciences, Allschwil, Switzerland) at 68ºC o/n

Filters were washed and exposed to a phosphor imager for 10 days (Molecular Dynamics, Sunnyvale, USA).

Samples corresponding to different time-points of recovery from exercise of one individual before or after training, respectively, were analyzed in parallel.

Expression was quantified from raw signal intensities in relation to the background as estimated using the Grid Background Dots mode for 54 selected dots (AIDA Array Easy software, Raytest, Urdorf, Switzerland).
