while under isoflurane anesthesia a unilateral full-thickness VML injury was created using a 6 mm punch biopsy to the middle third of the TA muscle. On average, the tissue removed from the TA weighed 90.5 ± 3.2 mg.

The muscles were thawed, minced and homogenized (Tissue Ruptor, Qiagen) for 30 seconds at room temperature and total RNA was isolated from the slurry using the miRNeasy Mini Kit (Qiagen) as per the manufacturer’s instructions. 1 μg of isolated total RNA was used to produce cDNA libraries using the Truseq protocol (Illumina), as per the manufacturer’s instructions. Biological replicates or triplicates were produced from each condition and individual libraries were pooled and sequenced using 76 base-pair (bp) paired-end reads on a Hi-Seq. 2500 in high output mode to an average depth of 55 M reads per library.
