Extraction protocol |
Placental tissue was collected from each placenta from four random sampling sites on the villous aspect. Placentas were then snap frozen in liquid nitrogen, homogenized and pooled to provide a sample of the entire placenta. Placentae obtained prior to 18 weeks gestation were identified based on their villous structure. The entire placenta was frozen and subsequently homogenized for total RNA extraction with a miRNeasy kit.

100 ng RNA was labelled using the miRNA Microarray System with miRNA Complete Labeling and Hyb kit Protocol Version 3.0.

Hybridization protocol |
Samples were hybridised to arrays for 20 hours at 55C rotating at 20 rpm.

Scan protocol |
Slides were scanned using the AgilentG3_miRNA scanner protocol for G3 format.

Data was extracted using Feature Extraction Software – FE protocol miRNA_QCMT_Jan09
