Total RNA was extracted from thymus blocks using mirVana™ miRNA isolation kit (Ambion) according to the manufacturer’s protocol. The RNA samples were treated with RNase Free DNase (QIAGEN, Valencia, CA) and were purified using RNeasy mini kit (QIAGEN) according to the manufacturer’s protocol.

Labeled samples with eukaryotic hybridization control were hybridized to GeneChip® miRNA 4.0 Array (Affymetrix) chips in a Gene Chip hybridization Oven 640 (Affymetrix) for 18hrs.
