Treatment protocol |
We generated il7r-/- homozygous mutant by clustered regularly interspaced short palindromic repeats (CRISPR) / Cas9 technology. Tu and il7r-/- larvae at 4dpf were collected and rinsed with E3 medium . They were put in 1.5 mL RNase tube after discarding excess water and stored in liquid nitrogen.

Growth protocol |
Zebrafish larvae were raised at 28.5°C under a 10/14h dark/light cycle . Embryos were collected after natural spawning and staged developmentally by hours post fertilization (hpf) or days post fertilization (dpf).

Trizol extraction of total RNA was performed according to the manufacturer's instructions.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on on Zebrafish Gene 1.0 ST Array. GeneChips were washed and stained in GeneChip Fluidics Station 450.

Scan protocol |
GeneChips were scanned using the GeneChip2 Scanner 30007G (Affymetrix).

Data processing |
The data were analyzed with RMA using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 100.
