Treatment protocol Ovariectomized mice were injected intraperitoneally with vehicle (100 ul saline) , E2 (250 ng in 100 ul saline) or IGF1 (50 ug in 100 ul PBS), or had an osmotic pump (Alza 100 3D; 3 day) containg IGF1 (0.5 mg/ml 0.1N acetic acid) inserted intraperitoneally. Uterine tissue was collected after 2 hours or 24 hours, frozen in liquid nitrogen and crushed
Extracted molecule total RNA
Extraction protocol RNA was isolated using Trizol reagent (Thermo Scientific). RNA was further purified using Qiagen RNAeasy RNA clean-up protocol with DNAse treatment
Label biotin
Label protocol Three hundred nanograms (300 ng) of total RNA were amplified and labeled as directed using the Affymetrix WT Plus Reagent Kit (WT Plus Kit).
 
Hybridization protocol Five and one half micrograms (5.5 μg) of amplified biotin-cDNAs were fragmented and hybridized to each array for 16 hours at 45°C in a rotating hybridization oven. Array slides were stained with streptavidin/phycoerythrin utilizing a double-antibody staining procedure and then washed for antibody amplification according to the GeneChip Hybridization, Wash and Stain Kit and user manual following protocol FS450-0001.
Scan protocol Arrays were scanned in an Affymetrix Scanner 3000.

Data processing Data was obtained using the GeneChip® Command Console software.
