Treatment protocol Human cell lines were treated with vitamin C in culture for 0, 12 and 72hrs. Mouse progenitors were treated with 1ug/ml Doxycycline to turn off Tet2 shRNA expression for 14 days

Data processing Base calling and demultiplexing was performed using Illumina bcl2fastq software.

The trimmed reads were aligned to the human and mouse genomes (build hg19/GRCh37 and mm10/GRCm38, respectively) using Bismark aligner.
Methylation calls were performed by Bismark.
