RNA of the rat heart was isolated using a mirVana miRNA isolation kit (Ambion, Austin, TX) and was reverse-transcribed using the miRNA reverse transcription kit in combination with stem-loop Megaplex primer pool (Applied Biosystems, Foster City, CA).

The PCR was run in a 7900HT Fast Real-Time PCR System (Applied Biosystems, Foster City, CA), and miRNA expression was calculated by comparative delta Ct analysis. U6 snRNA (Applied Biosystems, Foster City, CA) was used as an endogenous control.

Data processing |
The normalization and all the data analysis were performed according to the manufacturers instructions. For the normalization it uses the value of U6, a housekeeping gene. Target miRNA signals normalized to the housekeeping genes 2^-deltaCt, where deltaCt = (Ct_Target − Ct_HKG)] The web-based software package automatically performs all deltadeltaCt based fold-change calculations from the uploaded raw threshold cycle data. normalized data file reports normalized signal (against housekeeping genes). Fold Change file reports test/control (i.e., Diabetes/Non-diabetes) ratios.
