Sample GSM2536413 Query DataSets for GSM2536413
Status Public on Oct 11, 2017
Title IL-36β stimuated keratinocytes rep3
Sample type SRA
Source name Keratinocytes
Organism Homo sapiens
Characteristics cell type: keratinocytes
agent: IL-36β
passages: passage 2
Treatment protocol Keratinocytes were treated with 20ng/ml IL-36α, IL-36β, IL-36γ (Bio-Techne) or vehicle control. Cells were harvested after 24h.
Growth protocol Primary keratinocyte cultures were established as described elsewhere (Mahil et al, J Invest Dermatol 2016) using discarded healthy skin from a donor undergoing a plastic surgery procedure at Guy’s and St Thomas’ NHS Foundation Trust. Keratinocytes were cultured in Epilife medium containing Supplement 7 and 1% penicillin-streptomycin (all reagents Gibco).
Extracted molecule polyA RNA
Extraction protocol Total RNA was isolated using RNeasy Plus Mini Kit (Qiagen)
RNAseq libraries were prepared with the Illumina TruSeq Stranded Total RNA preparation kit and sequenced on an Illumina HiSeq3000 system.
Library strategy RNA-Seq
Library source transcriptomic
Library selection cDNA
Instrument model Illumina HiSeq 3000
Description B20_3
Data processing Sequenced reads were aligned to the Ensembl Human GRCh37 transcriptome using Bowtie 2-2.2.2. The number of reads that mapped to each gene was quantified with htseq-count.
Genome_build: GRCh37
