Treatment protocol |
Mouse trophoblast stem cells were cultured in the same media in the ‘growth protocol’ except 400mM sorbitol was added to cause hyperosmolar stress. Cells were harvested for analysis by microarray at 5 time points from 0-24hr of stress.

Growth protocol |
Mouse placental trophoblast stem cells were cultured in standard media supplemented with conditioned media from mouse embryonic fibroblasts and 10ng/ml fibroblast growth factor (FGF)4 and heparin. The latter two are known to keep trophoblast stem cells in a undifferentiated pluripotent state.

Extraction protocol |
Total RNA was isolated from cells 2-3 times for each time point. Trizol extraction of total RNA was performed according to the manufacturer's instructions.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on Murine Genome 430.2 Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.

Scan protocol |
GeneChips were scanned using the Affymatrix gene chip scanner 3000.
