Root samples were ground in liquid nitrogen and the RNA was extracted using the Qiagen Plant RNeasy kit. The RNA was then precipitated from the eluate with isopropanol

Approximately 5ug of purified total RNA was processed to produce biotinylated cRNA targets followed by streptavidin-phycoerythrin fluorescent staining according to the standard Affymetrix 3’-IVT labeling protocol
