10ng of inducible mRNA construct was injected into one cell embryos at around 20 degrees off the animal pole.

Growth protocol |
Embryos were injected with 10ng of each mRNA at the one cell stage and allowed to reach stage 9, whereupon animal caps were excised and cultured in .75 NAM for 2.5 hours in the presence of cycloheximide and dexamethasone.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 10 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip Xenopus leavis Genome Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.
