Title |
HTI-286 in docetaxel-refractory prostate cancer (Dye Flip 1) |
Sample type |
RNA |
Channel 1 |
Source name |
HTI-286 (Dye Flip 1)
Organism |
Homo sapiens |
Characteristics |
LNCaP prostate cancer cells
Biomaterial provider |
Dr. Leland W.K. Chung (Emory University, Atlanta)
Treatment protocol |
LNCaP cells were treated with HTI-286 (0.75nM) for 72h
Extracted molecule |
total RNA |
Extraction protocol |
Total RNA was isolated using the Trizol method (Invitrogen) according to manufacturer’s protocol. RNA quality was assessed with an Agilent 2100 Bioanalyzer (Agilent Technologies, Mississauga, ON, Canada) and RNA 6000 nano kit (Agilent). RNA was quantitated using the Nanodrop ND-1000 Spectrophotometer, Nanodrop Technologies, Wilmington, DE
Label |
Cy5
Label protocol |
10ug of total RNA was used to generate Cy5-labeled 3DNA DNA Dendrimer (Genisphere, Hatfield, PA) probes according to the manufacturer’s (Genisphere, Hatfield, PA) directions and hybridized to 70-mer oligonucleotide arrays.

Label |
Cy3
Label protocol |
10ug of total RNA was used to generate Cy3-labeled 3DNA DNA Dendrimer (Genisphere, Hatfield, PA) probes according to the manufacturer’s (Genisphere, Hatfield, PA) directions and hybridized to 70-mer oligonucleotide arrays.
Hybridization protocol |
3DNA array detection 350 kit (Genisphere, Hatfield, PA)
Scan protocol |
Two fluorescent images (Cy3 and cy5) were scanned separately using a Perkin Elmer ScanArray Express Scanner (PerkinElmer Life and Analytical Sciences, Inc., Wellesley, MA)
Description |
HTI-286 in docetaxel-refractory prostate cancer
Data processing |
Arrays were scanned at excitation wavelengths of 532 and 632 nm to detect Cy3 and Cy5 respectively. Signal quality and quantity were assessed using Imagene 5.6 (Biodiscovery, San Diego, CA).Signal median from Imagene were analyzed in GeneSpring 6.1 (Silicon Genetics, Redwood City, CA)
