At surgery, right adrenal tumor and lymph node metastasis were resected. Then, small peccieces of primary tumor was picked up and frozen immediately until use.

Total RNA was extracted from tissue specimen using RNA isolation kit according to the manufacturer’s instructions (Stratagene, Agilent techonology, Santa Clara , CA) and purified further using the Rneasy Mini Kit (Invitrogen).

Conversion of double stranded cDNA into biotin-labeled cRNA was accomplished using the One-Cycle cDNA Synthesis Kit (T7) (Affymetrix, Santa Clara, CA) according to the manufacturer’s instructions.

After hybridization cocktails preparation, the Human Genome U133 Plus 2.0 Array was hybridized with the fragmented labelled cRNA for 16 h at 45 °C as described in the Affymetrix Technical Analysis Manual.

Scan protocol GeneChips, bound to streptavidin-phycoerythrin, were processed using the Affymetrix GeneChip Fluidics Station 450 and scanned with an GeneChip Scanner 3000 (Affymetrix).
