Title |
GFP_cntl_3 |
Sample type |
RNA |
Source name |
lentivirus-transduced HFF cells
Organism |
Homo sapiens |
Characteristics |
cell type: HFF cells lentivirus transduction: GFP-control construct time: 5d-post-transduction
Treatment protocol |
HCMV infections were performed as described in Stark et al., 2012 and Belzile et al., 2014. For lentivirus transductions (GFP and CPEB expression cells), puromycin drug selection was initiated at 48 h, and cells were harvested 3 days subsequently.
Growth protocol |
HFFs and ES-derived NPCs were cultured as described in Sanders et al., 2008 and in Belzile et al., 2014.
Extracted molecule |
total RNA |
Extraction protocol |
TRIzol (Invitrogen/Life Technologies) extraction of RNA, according to the manufacturer’s instructions.
Label |
biotin
Label protocol |
WT Terminal Labeling Kit, Affymetrix
Hybridization protocol |
Hybridization cocktails containing ~5.5 ug of fragmented and labeled DNA target were prepared and applied to GeneChip Human Splice Junction arrays. Hybridization was performed for 16 hours using the Fluidics 450 station.
Scan protocol |
Arrays were scanned using the Affymetrix 3000 7G scanner and GeneChip Operating Software version 1.4 to produce .CEL intensity files.
Description |
Array Sample 15
Data processing |
Data processed using Affymetrix package (Affy Power Tools) apt-probeset-summarize. Iter-plier algorithm used to quantify probesets. As previously described (Huelga et al., 2012). HJAY_r2.pgf
