Adoptively transferred OT-I CD8 T cells were primed in vivo with SIINFEKL peptide and CD134 plus CD137 costimulatory agonists and FACS sorted from spleens 4 days later and then treated in vitro with or without 0.5 ug/ml IL-2 for 1 hour

Treated cells were snap frozen and shipped to ActiveMotif, Inc. who performed the STAT5A ChIP-seq analysis according to their FactorPath™ assay.

The 50-nt sequence reads identified by the Sequencing Service (using Illumina’s Hi-Seq) are mapped to the genome using the BWA algorithm with default settings. Alignment information for each read is stored in the BAM format. Only reads that pass Illumina’s purity filter, align with no more than 2 mismatches, and map uniquely to the genome are used in the subsequent analysis. In addition, unless stated otherwise, duplicate reads (“PCR duplicates”) are Genome_build: removed. Approximately 17 million non-duplicate reads per sample were alligned to the mouse genome (mm10).
