Growth protocol |
CD14+ cells from bone marrow of 8 MGUS, 11 SMM and 16 MM patients. The CD14+ cell fraction was obtained by immunomagnetic cells sorting using anti-CD14 monoclonal antibody-coated microbeads (MACS, Miltenyi Biotec., Bergish-Gladbach, Germany). Samples with a purity of more than 90%, tested via flow-cytometry (BD biosciences, San Josè, CA) were analyzed. These samples were evaluated immediately after purification.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 5 micrograms of total RNA (Expression Analysis Technical Manual, Affymetrix).

Hybridization protocol |
Following fragmentation, 15 micrograms of cRNA were hybridized for 16 hr at 45°C on GeneChip HG-U133 Plus 2.0 Arrays. GeneChips were washed and stained in the Affymetrix Fluidics Station 450.

Scan protocol |
HG-U133 2.0 Plus arrays were scanned using the Scanner G3000 7G (Affymetrix).

Data processing |
The probe-level signals were converted to expression values using the Bioconductor function for Robust Multi-array Analysis (RMA), in which perfect match intensities are background adjusted, normalized by means of quantile-quantile normalization and log2-transformed.
