Treatment protocol |
LNCap and VCap cells were added 10nM OBP and incubated for 48 hours at 37 ºC in a humidified incubator with 5% CO2.

Growth protocol |
LNCap and VCap cell lines were purchased from ATCC (Manassas, VA, USA). These cells were cultured in RPMI 1640 (Life Technologies, Carlsbad, CA) supplemented with 10% heat-inactivated fetal bovine serum at 37 ºC in a humidified incubator with 5% CO2.

Extraction protocol |
Total RNA was isolated using TRIzol (Life Technologies) following the manufacturer's recommendations. RNA was quantified using a NanoDrop-2000.

One hundred ng of total RNA was labeled with pCp-Cy3 (Agilent Technologies) and 15 units of T4 RNA ligase (GE Healthcare, Little Chalfont, Buckinghamshire, UK) at 16°C for 2 hours.

Hybridization protocol |
Labeled samples were purified with MicroBio-Spin 6 columns (Bio-Rad, Richmond, CA) and hybridized to microarray at 55 °C with rotation at 20 rpm for 20 hours.

Scan protocol |
Slides were scanned immediately after washing on the Agilent DNA Microarray Scanner (G2505C) using one color scan setting for 8x60k array slides.

Data processing |
The scanned images were analyzed with Feature Extraction Software 10.7.1.1 (Agilent) using default parameters (protocol miRNA_107_Sep09) to obtain background subtracted and spatially detrended Processed Signal intensities. Features flagged in Feature Extraction as Feature Non-uniform outliers were excluded.
