Human samples of cerebral aneurysm and superficial temporal arteries were extracted surgically from patients undergoing craniotomy for cerebral aneurysms.

Extraction protocol |
After successful clipping of the aneurysm, the dome was resected with the use of micro-scissors and bayonetted forceps (not used before during the case) and was placed immediately into a vial containing RNAlater solution (Ambion, Austin, TX) to protect RNA from degradation. The vials were immediately frozen to -80 °C. Small RNA was then size selected with mirVana miRNA isolation kit.

Following the manufacturer’s protocol, 130ng of total RNA was labeled with the FlashTag Biotin HSR RNA labeling kit (Affymetrix, Santa Clara, CA). To confirm efficient labeling the ELOSA QC assay (Affymetrix, Santa Clara, CA) was performed.

Hybridization protocol |
Samples were hybridized on the Affymetrix 4.0 microarrays overnight.

Scan protocol |
Washing, staining and scanning of the arrays was performed using the Command Console, and the spike-in analysis of control oligonucleotides was evaluated using the Expression Console.

Data processing |
Data were analyzed using the recommended Transcriptome Analysis Console (TAC) software from Affymetrix. Only human microRNAs were included in the analysis, resulting in reduced data set of 6,631 rows.
