Treatment protocol |
Whole mouse lung tissues were homogenized in liquid nitrogen. Total RNAs were extracted from mouse lungs using QIAGEN RNeasy Mini Kit.

Growth protocol |
Mice were anesthetized with ether and were intranasally inoculated with 104 TCID50 of PR8 virus in physiological saline.For the total RNAs extraction, entire lungs from mice infected by PR8 virus were randomly selected and harvested on the 2th and 5th day.

Biotinylated cRNA were prepared according to the standard Affymetrix protocol from 6 ug total RNA (Expression Analysis Technical Manual, 2001, Affymetrix).

Hybridization protocol |
Following fragmentation, 10 ug of cRNA were hybridized for 16 hr at 45C on GeneChip miRNA 4.0 Array. GeneChips were washed and stained in the Affymetrix Fluidics Station 400.

Scan protocol |
GeneChips were scanned using the Hewlett-Packard GeneArray Scanner G3000 7G.

Data processing |
The data were analyzed with RMA using Affymetrix default analysis settings and global scaling as normalization method. The trimmed mean target intensity of each array was arbitrarily set to 100.
