A27 bone marrow aspirate was collected with informed consent. after red blood cells lysis, nucleated cells were counted and 4.7x10(5) cells per square centimeter were plated. cells were cultured in T75 flask (Becton Dickinson) containing 30mL of a growth medium consisting of a-MEM, 2mM L-glutamine, 100U/mL penicillin, 20% fetal bovine serum (FBS, Invitrogen). medium was replaced on day 1 and then twice a week. first passage was performed on days 12-16 when colonies began to merge and cells were replated in T75 flasks at 0.1x10(5) cells/cm(2).
