Treatment protocol |
Primary hepatocytes were transduced with non-specific shRNA, shKAT2B or shWDR5 (2-4 pfu/cell) and cultured for 72 hours. Cells treated with 100nM glucagon or PBS for 90 min.

Growth protocol |
Primary hepatocytes were grown in Medium 199 from Mediatech supplemented with 100mg/ml Penicillin/Streptomycin. Medium was changed daily.

Extraction protocol |
Total RNA was extracted using Trizol followed by the Qiagen RNAeasy kit according to manufacturer's instructions. Each sample contains RNA from three separate culture dishes.
