Title |
Sox2 is required for global chromatin interactions in brain-derived neural stem cells [ChIP-seq] |
Organism |
Mus musculus |
Experiment type |
Genome binding/occupancy profiling by high throughput sequencing
|
Summary |
Genome wide mapping of Sox2 binding sites in neural progenitor cells
|
|
|
Overall design |
ChIP-Seq experiment on transcription factor Sox2 in neural progenitor cells. Cells were collected as small neurospheres and fixed either directly ("Spheres") or after dissociation ("Singles"). Cells were fixed sequentially with di(N-succimidyl) glutarate and 1% formaldehyde in phosphate-buffered saline and then lysed, sonicated and immunoprecipitated as described previously (Mateo et al., 2015 and references therein). SOX2 was immunoprecipitated with 3mg of goat anti-SOX2 (Santa Cruz sc-17320).
