Title |
Global gene expression pattern in Mouse Trophoblast Stem Cells (mTS) with or without knocking down of Tead4 |
Organism |
Mus musculus |
Experiment type |
Expression profiling by high throughput sequencing
|
Summary |
A TEA domain containing transcription enhancer factor, TEAD4 (TEF3) was found to be very important in the trophectoderm (at preimpantation embryos in rodent) in some earlier studies (Home et al, Saha et al, 2012). Here we report the role of TEAD4 in the TE derived mTS cells with a loss of function approach where the mTS cells under stem condition were transduced with lentiviral shRNA construct and the global gene expression was analysed with the help of a third generation sequencer.

Overall design |
mTS cells under stem condition was transduced with lentiviral particles consists of scramble or shRNA against Tead4. The puromycin selected cells were passaged and then the whole cell-lysates were used to prepare RNA. RNA-seq analysis were performed using a illumina HiSeq2500 platform.
