Summary |
The hpCasMINI exhibits high specificity for gene editing. Compared to SpCas9 and LbCas12a, hpCasMINI also exhibited higher specificity for genomic DNA cleavage at the tested sites.

Overall design |
We evaluated the specificity of hpCasMINI-mediated gene editing in human cells using Tag-seq. Within the 10 tested sites, hpCasMINI exhibited about 2.5-fold increased activity and similar specificity compared to CasMINI. we also compared the DNA cleavage activity of SpCas9, LbCas12a, CasMINI, and hpCasMINI across four genomic sites at the same location and five sites at adjacent locations, hpCasMINI also exhibited higher specificity for genomic DNA cleavage at the tested sites.
