Overall design |
Blood was obtained from NOD mice via retro-orbital bleeding. Blood was then ficolled and PBMCs were isolated. CD4 T cells were then single cell sorted from PBMCs via MHC tetramers. Macrophages and Endothelial cells were isolated from pancreatic islets. In short, pancraetic islets were isolated form NOD pancreas via tissue harvesting and processing via enzymatic digestion and ficoll. Cells were either sorted with pMHC tetramers or antibody surface staining. There is no processed data file because all analysis was completed using custom R scripts and there is no need for normalization of our data.
