Summary |
To understand the impact of ANCO1 reduction on gene expressions, we performed RNA-seq experiments in shANCO1 and shCTRL MCF10A and MCFDCIS cells in 2D and 3D culture. We report ANCO1 knockdown leads to altered gene expression profiles. Enriched gene sets including chr1q21.3 and mTORC1 signaling may be responsible for phenotypical changes in shANCO1 cell lines.

Overall design |
RNA-seq experiments were performed in 2D- and 3D-cultured shANCO1 and shCTRL MCF10A and MCFDCIS cell lines to examine changes in transcriptome. At least two shANCO1 constructs targeting different regions of ANCO1 were used in each setting. Experiments were done in triplicates.
