Title MPA induced alterations in endometrial stromal cell transcriptome profile

Summary Normal endometrial stromal cells (ESCs) increase the inhibitory effect of progestin on EC cell proliferation via paracrine signaling, but the mechanisms involved remain unclear.ESCs had different morphological features between progestin-sensitive and -insensitive EC tissues. The purpose of this study was to detect differential gene expression in endometrial stromal cells after MPA treatment and to screen for potential secretory factors.
 
Overall design The NSC cells were digested and passaged routinely at logarithmic growth stage, and then inoculated in six-well plates at 1*105/well after adjusted cell concentration. After 24h of cell walling, the cells were observed to be 60%-70% confluent, and the phenol red-free medium was starved for 8h, then replaced with phenol red-free medium supplemented with 2% charcoal-stripped FBS, and treated with ethanol or 10μM MPA for 6h, and then Trizol method was used to extract RNA. RNA-Seq was performed by Genergy Bio-Technology (Shanghai, China).
