Summary |
We examined the transcriptional and translational response to BRAF targeted therapy in BRAF mutant melanoma by applying RNA-seq to polysome profiling, a method that isolates mRNA bound to actively translating ribosomes. By applying a published data analysis pipeline, anota2seq, we determined differentially expressed genes in total RNA and polysome-bound RNAseq datasets. We performed the analysis on two treatment timepoints, allowing determination of changes in expression that occur across different treatment times. These analyses identified extensive translational reprogramming during the BRAF targeted therapy response.
