Title Profiling the transcriptional and translational response to BRAF targeted therapy in melanoma
Organism Homo sapiens
Experiment type Expression profiling by high throughput sequencing
Summary We examined the transcriptional and translational response to BRAF targeted therapy in BRAF mutant melanoma by applying RNA-seq to polysome profiling, a method that isolates mRNA bound to actively translating ribosomes. By applying a published data analysis pipeline, anota2seq, we determined differentially expressed genes in total RNA and polysome-bound RNAseq datasets. We performed the analysis on two treatment timepoints, allowing determination of changes in expression that occur across different treatment times. These analyses identified extensive translational reprogramming during the BRAF targeted therapy response.

Overall design Examination of total mRNA and polysome-bound mRNA in BRAF mutant melanoma cells treated with the BRAF inhibitor vemurafenib (0, 24h, 40h).
