Title RNA seq analysis of MC38CEA cells transduced with pLKO1 vectors encoding shRNAs targeting Adam17 or control shRNA

Summary The goal of this study was to compare transcriptomes of cells with reduced and normal levels of ADAM17, unstimulated or stimulated with IL1b and IFNg
 
Overall design The cells were transduced with pLKO vectors from MISSION® library encoding shRNA targeting mouse ADAM17: SL2 (TRCN0000031953) or control non-targeting shRNA: SHC016. Transduced cells were selected with puromycin. The cells were left unstimulated or were stimulated with human recombinant (hr) IL1b (10 ng/ml) and mouse recombinant (mr) IFNg (10 ng/ml) for six hours and RNA was isolated. Note: Control shRNA, SHC016, is cytotoxic and its expression leads to elimination of shRNA-coding casette (personal observation).
