Overall design |
B-cells isolated from 7 COVID-19 patient's peripheral blood were seperated into ACE2 reactive and non-reactive population using ACE2 peptides. Each of these patient's B-cells were labelled with Hashtag oligo linked to antibody, which allowed to barcode each patient with unique DNA oligo sequence. Then ACE2 reactive B cells from 7 patients were pooled together and loaded on 10x Genomics Chip G using 5'DGE kit to interogate single cell 5DGE (Digital gene expression) and VDJ sequence in B-cells. Similarly, ACE2 non-reactive cells were pooled together.
