Summary |
We performed RNA-seq and ATAC-seq on UCs from healthy humans and human embryonic stem cell line H1. By analyzing the transcriptome expression profile and chromatin accessibility bettween them, we find new transcription factors that can be used for reprogramming to study the mechanism of reprogramming.

Overall design |
RNA-Seq analysis was performed on different groups of cells on the last day of reprogramming (D16) to try to find the specific role of TEAD2\TEAD4. The cell lines reprogrammed with tead2, tead4 and ZIC3 renamed as UiPSC-TEAD2, UiPSC-TEAD4 and UiPSC-ZIC3. Then we performed RNA-seq sequencing of UiPSC-TEAD2, UiPSC-TEAD4 and UiPSC-ZIC3, UiPSC, H1 and UC of different generations. UiPSC-TEAD2, UiPSC-TEAD4 and UiPSC-ZIC3 were found to have very similar expression profiles to UiPSC, and they are all similar to H1, but are quite different from UC. This study shows that Tead2/Tead4/Zic3 can promote UCs reprogramming.
