Summary |
ZFP541 is assumed to possess putative DNA-binding domains and associated with histone deacetylases, implying it plays a role in regulation of transcription via modulation of chromatin status. Therefore, we used ChIP-seq to search for targets of ZFP541 in meiotic prophase of mouse spermatocytes.

Overall design |
ZFP541 ChIP was performed using chromatin lysates, that were derived from wild type testes at postnatal day 18-19, and protein A Dyna-beads (Thermo-Fisher) coupled with 5 μg of rabbit anti-ZFP541-N antibodies (#1 and #2). ChIP-seq libraries were prepared using 20 ng of input DNA, and 4 ng of ZFP541 ChIP DNA with KAPA Library Preparation Kit (KAPA Biosystems) and NimbleGen SeqCap Adaptor Kit A or B (Roche) and sequenced by Illumina Hiseq 1500 to obtain single end 50 nt reads. ChIP-seq reads were trimmed to remove adapter sequence when converting to a fastq file.
