Title Single-cell lineage mapping of a diverse virus-specific naïve CD4 T cell repertoire

Summary The naïve polyclonal CD4 T cell repertoire can differentiate into multiple lineages during acute viral infection, with some T cell clones exhibiting a preferred lineage choice. TCR α CDR3 motifs can partially influence a clone-intrinsic lineage preference towards TFH fate.
Overall design Activated antigen specific CD4 T cells (CD4+CD44+GP66+) were sorted from five C57BL/6J mice and used to perform Single cell gene expression (GEX) and single cell TCR sequencing (TCR) using 10x genomics kit at day 10 post acute LCMV infection
