Expression profiling by high throughput sequencing
Summary
We performed RNA-seq on head tissue collected from Drosophila expressing N-terminal (UAS-HTTNT231Q128) or full-length (UAS-HTTFL200Q) human mHTT in neurons (elav-GAL4) or glia (repo-GAL4).
Overall design
For each combination of HD model and driver, RNA-seq was performed at three time points to capture the early, middle, and late phases of disease pathogenesis, corresponding to behavioral deficits caused by mHTT-induced neuronal or glial dysfunction. At each timepoint, samples for HD and age-matched controls were collected in triplicate.
