Summary |
Optimal Ang II levels during postnatal life are required for a normal maturation of the rat kidney. This study investigated both short- and long-term effect of MatSep on: 1) the status of intrarenal renin-angiotensin system components, 2) the isolated renal vasculature genome-wide gene expression trajectory from neonatal to adult life, and 3) the renal vascular architecture and function. In this dataset, gene expression changes by MatSep at P10 in renal vasculature isolated from neonates, modulate extracellular structure organization, vasculature development, inflammation and angiogenesis. The long -term effects of MatSep on renal vasculature gene expression is assessed at P180, which reveal clusters of genes with short, long, and downstream changes in expression.

Overall design |
20 total samples were analyzed in 4 experimental groups: un-stressed (control littermates) and stressed (MatSep) neonates (P10) and adult (P180) rats (n=5 per group). We performed pair-wise comparisons and defined Early-life genes as those with expression levels at P10 > at P180 in un-stressed controls and Adult-life genes as those with expression levels at P180 > at P10. Early life MatSep affected genes were identified by pair-comparison between P10 MatSep group and P10 controls. The long-term MatSep effects on expression of genes were identified by pair-comparison between P180 MatSep group and P180 controls.
