Summary |
Transmembrane adaptors on T cells play crucial roles in TCR signal transduction. Although the TCR-LAT signal-transduction axis occupies a central place in T cell activation, it does not work in isolation and is tuned by other T-cell surface receptors among which stand the CD5 and CD6 receptors. Here we assessed the extent of transcriptional changes that initiated by TCR-CD28 pathways in the absence of CD5, of CD6, of both CD5 and CD6, or of LAT by RNA-seq analysis.

Overall design |
RNA-seq analysis of untreated and stimulated naïve CD4+ T cells of wild-type, CD5 KO, CD6 KO, CD5/CD6 double KO and LAT KO. Purified T cells were stimulated with anti-CD3 plus anti-CD28 for 4 h and 20 h. WT (hDTR+) and Lat–/– (hDTR–) naïve CD4+ T cells were purified from pooled lymph nodes and spleens from Latfl-dtr maT-Cre mice using FACS AriaIII as described in Roncagalli et al (2014). There were 3 biological replicates for each conditions.
