Summary |
T cell factor 1 (Tcf1, encoded by Tcf7) promotes the central memory CD8+ T cell (TCM) differentiation and homeostatic proliferation in lymphoid tissues. It remains unclear if and how Tcf1 regulates the CD103high tissue-resident memory CD8+ T cell (TRM) formation in non-lymphoid tissues. Using an influenza A infection mouse model, we collected Tcf7-deficient and -sufficient lung CD8+ TRM cells for RNA sequencing analysis.

Overall design |
To understand the global gene expression profiles in Tcf7-deficient and -sufficient lung CD8+ TRM cells, we infected mice receiving Tcf7-deficient and -sufficient TCR transgenic P14 CD8+ T cells with influenza A virus X31 expressing LCMV epitope GP33-41 (X31-33). Eight days after infection, the mice were injected with PE-conjugated anti-CD8beta to intravascularly label circulating CD8+ T cells. Lung-infiltrating T cells were stained with CD8a and CD103. CD103high Tcf7-deficient and -sufficient P14 CD8+ T cells were FACS-sorted on day 8 after X31 infection for RNA extraction and RNA sequencing (RNA-seq) analysis.
