Summary |
To understand the role of YAP/TAZ during gliomagenesis, we compared the transcriptomes of control and YAP/TAZ knockout astrocytes derived from newborn mice, transformed with oncogenic HER2.

Overall design |
Yapf/f;Tazf/f mouse astrocyte were expanded and transduced with lentiviral particles encoding for doxy-inducible version of HER2CA, rtTA and CreERT2. Cells were also treated with DMSO (control) or TAM to promote YAP/TAZ knockout during the oncogene transformation. Not transformed astrocytes were used as control. Fully formed P1 Gliomaspheres were also profiled as comparison. After 15 days, cells were harvested for RNA extraction. Gliomaspheres were harvested for RNA extraction 3 weeks after the beginning of the experiment; RNA was then used for polyA RNA-sequencing.
