Expression profiling by high throughput sequencing
Summary
Obesity and type 2 diabetes (T2D) can be associated with altered secretion of enterohormones in condition that remains to be understood in depth. Here, we aimed to decipher the mechanisms by which a major enterohormone GLP-1, is decreased in human obese patients according to their diabetic status.
Overall design
Patient candidates for bariatric surgery were phenotyped and pieces of jejunum were gathered. Jejunum epithelial samples from non-obese, obese and obese-diabetic patients were collected. Samples were assayed for enteroendocrine cell (EEC) densities and RNA for transcription factors known to drive EEC lineage. EEC enrichment was performed and transcriptional profile was established by RNA sequencing to identify unknown factors in relation with GLP-1 production and secretion in another group of 14 obese and 13 obese-diabetic patients
