Title |
Single cell RNA sequencing of lung adenocarcinoma |
Organism |
Homo sapiens |
Experiment type |
Expression profiling by high throughput sequencing
|
Summary |
We performed single cell RNA sequencing (scRNA-seq) for 208,506 cells derived from 58 lung adenocarcinomas from 44 patients, which covers primary tumour, lymph node and brain metastases, and pleural effusion in addition to normal lung tissues and lymph nodes. The extensive single cell profiles depicted a complex cellular atlas of lung adenocarcinoma progression which includes cancer, stromal, and immune cells in the surrounding tumor microenvironments.

Overall design |
All single-cell mRNA expression profiles were acquired from forty-four patients pathologically diagnosed with lung adenocarcinoma (LUAD) (58 samples). 11 tumour, 11 distant normal lung, 10 normal lymph node, and 10 metastatic brain tissue samples were obtained from LUAD patients conserving surgery without prior treatment. 7 metastatic lymph node and 4 lung tumour tissue samples from advanced stage LUAD patients were also collected through the endobronchial ultrasound (EBUS) and bronchoscopy (BRONCHO). And 5 pleural fluid samples were obtained from LUAD patients with malignant pleural effusion. We applied the quality measures so as to remove less informative cells and finally obtained 208,506 single cell sequencing data.
